The series invites researchers and leaders in computational pathology and related fields to present their work and foster thought-provoking discussions. \nThis seminar features Andrew Zhang from Harvard Medical School\, Boston\, USA. \nAbstract\nThe field of medical AI has traditionally focused on isolated clinical domains\, such as chest X-rays or pathology slides. These data modalities have the advantage of being plentiful\, relatively standard in format\, and information dense. But by looking at only individual snapshots of a patient’s medical journey\, are we missing the forest for the trees? Outside of the world of curated medical imaging datasets\, there are petabytes of clinical data existing in the electronic health record (EHR) as unstructured or semi-structured longitudinal events. These include diagnoses\, symptoms\, lab tests\, clinical reports\, medications\, and more\, each tied to a specific timepoint in the course of a patient’s healthcare journey. With our capacity to train increasingly larger and more powerful AI models\, the next frontier is to build a virtual patient: a model that understands and makes predictions on the entire EHR. In this talk\, I will introduce APOLLO\, a major effort towards modeling whole patients: advancing the state of the art not only in data size (25 billion events from 7.2 million patients) but also in data breadth (structured data\, clinical text\, and images) and downstream capabilities. To evaluate the potential of this model\, we created a large-scale benchmark consisting of over 300 patient retrieval and clinical forecasting tasks and showed that APOLLO patient embeddings generalize across medical domains and encode both retrospective and prognostic information about each patient. APOLLO has exciting applications not only in disease risk prediction but also as a multimodal medical search engine and unlocks the possibility of biomarker discovery from the entire EHR. \nRegister NowBio\nAndrew Zhang is a 4th-year PhD student in the Harvard-MIT Health Sciences and Technology Program\, working in the lab of Dr. Faisal Mahmood at Harvard Medical School. Prior to his PhD\, Andrew studied cell biology and computer science at Harvard. His work centers on advancing self-supervised representation learning at multiple biological and temporal scales: from encoding a single pathology slide to encoding entire longitudinal medical histories.

The series invites researchers and leaders in computational pathology and related fields to present their work and foster thought-provoking discussions. \nThis seminar features Dr Junlin Hou from The Hong Kong University of Science and Technology\, Hong Kong. \nAbstract\nComputational pathology is rapidly moving from task-specific image analysis toward foundation models that enable scalable\, generalizable disease diagnosis and clinical decision support. In this talk\, I will first present our recent work\, PathSegmentor\, a foundation model that enables flexible\, text-prompted segmentation across diverse pathological entities. Beyond segmentation\, I will also provide an overview of our broader efforts in pathology foundation models\, including image enhancement\, whole-slide representation learning\, benchmarking\, and agentic AI systems. These works aim to advance computational pathology toward more precise\, interpretable\, and clinically useful AI for precision oncology. \nRegister NowBio\nDr. Junlin Hou is a Post-doctoral Fellow in the Department of Computer Science and Engineering at The Hong Kong University of Science and Technology (HKUST). She obtained her Ph.D. from the School of Computer Science at Fudan University in 2023. She has published 30+ papers in top-tiered journals and conferences\, including Nature Computational Science\, Nature Biomedical Engineering\, IEEE TMI\, MICCAI\, CVPR\, etc. She has served as a guest editor for CMIG and J. Imaging\, and as a reviewer for 15+ journals and conferences. She also led the team winning 10+ medical grand challenges. Her current research interest is in trustworthy medical AI and computational pathology.

SUMMARY:How Close Are We to Personalized Treatment Decisions with Causal Machine Learning?

The series invites researchers and leaders in computational pathology and related fields to present their work and foster thought-provoking discussions. \nThis seminar features Octavia Ciora and Maresa Schröder from LMU Munich\, Munich\, Germany. \nAbstract\nUnderstanding the efficacy and safety of treatments is crucial for optimizing patient outcomes. However\, patients respond differently to the same treatment due to biological\, clinical\, or contextual factors\, which highlights the need for personalized clinical evidence to guide treatment decisions. Causal machine learning offers a promising way to do so by combining causal inference principles with flexible ML models to estimate personalized treatment effects. In this talk\, we present findings from a scoping review of 70 clinical studies applying causal machine learning in personalized medicine. We assessed 41 key methodological choices\, including clinical context\, model\, validation\, heterogeneity analysis\, and clinical translation. We show how these causal ML is currently used\, how their results are validated\, and where important gaps remain. Finally\, we propose a validation framework with methodological recommendations to promote reliable clinical evidence generation. \nRegister NowBio\nOctavia Ciora is a second-year PhD student in computer science at LMU Munich\, supervised by Prof. Stefan Feuerriegel. She holds bachelor’s and master’s degrees in bioinformatics from the Technical University of Munich and LMU Munich. Her research lies at the intersection of causal machine learning and healthcare. Her work focuses on developing machine learning-based frameworks for clinical decision-support\, treatment effect estimation from electronic health records\, and personalized treatment recommendations for oncology. Maresa Schröder is a final-year PhD student in computer science at LMU Munich\, supervised by Prof. Stefan Feuerriegel. She holds a mathematics master’s degree from the Technical University of Munich and bachelor’s degree in economics and mathematics from the University of Mannheim. Her research interest lies in causal machine learning with a special focus on reliable treatment effect estimation\, such as the development of uncertainty quantification and robust estimation methods while ensuring ethical constraints. Furthermore\, she is interested in developing advanced causal methods for medical applications and strategic decision-making.

She did her postdoctoral training and worked later as a staff scientist in the Laboratory of Cancer Biology and Genetics\, NCI. Her research focused on identifying mechanisms involved in the differential effects of natural compounds and their engineered derivatives acting on protein kinase Cs (PKCs)\, validated targets for anti-cancer treatment. In 2018 she joined the Collaborative Protein Technology Resources (OSTR\, CCR\, NCI) core facility to implement a variety of spatial protein and transcriptomics technologies\, and lead related operations as services to NCI investigators. Noemi provided expertise for development of project based customized antibody protein panels for detection of 40-60 protein targets at single cell level in tissue sections using the CODEX/Phenocycler fusion technology\, and ensured optimal and efficient instrument use. In addition\, she advised and supported users on tools for quantitative image analysis combining different AI-enabled HALO modules and NCI developed pipelines for downstream analysis. Noemi joined Indica Labs as Director of Product – Life Sciences in May\, 2025.

SUMMARY:TIA Centre Seminar Series – Co-evolving Computational Pathology: From Interactive Foundation Models (VISTA-PATH) to Agentic Medical Image Analysis (TissueLab)

The series invites researchers and leaders in computational pathology and related fields to present their work and foster thought-provoking discussions. \nThis seminar features Dr Peixian Liang and Songhao Li from the University of Pennsylvania\, USA. \nAbstract\nComputational pathology is rapidly moving beyond static\, single-task predictors toward interactive systems that can segment\, reason\, and quantify tissue morphology under expert supervision. In this talk\, we present two complementary pillars of this transition from our group. First\, we introduce VISTA-PATH\, an interactive foundation model for histopathology image segmentation designed to resolve heterogeneous tissue structures\, incorporate expert feedback\, and generate pixel-level segmentations that are directly meaningful for clinical interpretation. Trained on over 1.6 million image–mask–text triplets spanning 9 organs and 93 tissue classes\, VISTA-PATH outperforms existing segmentation foundation models and enables clinically relevant biomarkers\, such as the Tumor Interaction Score (TIS)\, which shows a strong association with patient survival. Second\, we present TissueLab\, an open\, co-evolving agentic system (tissuelab.org) for end-to-end medical image analysis. TissueLab unifies foundation models and analytical tools within a modular tool graph\, including VISTA-PATH for interactive segmentation\, and translates LLM-generated plans into executable operations. Through tool-level\, strategy-level\, and workflow-level co-evolution\, it achieves over 90% accuracy in cancer cell quantification within 10-30 minutes of expert feedback\, improves lymph-node metastasis correlation from 0.827 to 0.933\, and reaches a macro-AUC of 0.838 on tubule formation scoring\, outperforming human-orchestrated workflows and state-of-the-art VLMs. \nRegister NowBio\nPeixian Liang is a Postdoctoral Researcher at the University of Pennsylvania\, working with Prof. Zhi Huang. She received her Ph.D. degree in Computer Science from the University of Notre Dame\, Notre Dame\, IN\, USA\, in 2023. Her research focuses on developing innovative AI/ML methods to advance medicine. She is particularly interested in building foundation models and computational frameworks to assist pathologists\, reduce variability in diagnosis\, and identify clinically relevant morphologic biomarkers. Her work aims to advance AI-driven approaches for more precise diagnosis\, prognosis\, and clinical and biological discovery. Songhao Li is a first-year Ph.D. student in Electrical and Systems Engineering at the University of Pennsylvania\, advised by Prof. Zhi Huang. He received his M.S. in Computer Science from New York University in 2025 and brings a strong full-stack and system-engineering background from prior roles in industry. His research focuses on agentic AI\, human–AI collaboration\, and ML/DL for medicine\, aiming to bridge AI research and clinical practice through transparent\, interactive\, and continuously learning systems.

SUMMARY:TIA Centre Seminar Series – Computational Pathology for Discovering Biomarkers of Immunotherapy Response in Advanced Melanoma

The series invites researchers and leaders in computational pathology and related fields to present their work and foster thought-provoking discussions. \nThis seminar features Dr Mark Schuiveling from UMC Utrecht\, the Netherlands. \nAbstract\nImmune checkpoint inhibition (ICI) has significantly improved outcomes for patients with advanced cutaneous melanoma\, leading to durable responses and potential long-term remission in approximately half of patients. However\, it is also associated with severe and sometimes irreversible toxicity\, underscoring the need for reliable biomarkers. In our group\, we investigate whether computational pathology approaches add value beyond clinical data for identifying biomarkers associated with treatment response in these patients. In this talk\, I will highlight our work on tumor-infiltrating lymphocytes in melanoma histopathology\, as well as our recent work in the PUMA challenge\, where we assessed whether additional immune cell subsets provide further associated predictive value. \nRegister NowBio\nMark Schuiveling is a medical doctor and PhD candidate at the University Medical Centre Utrecht\, the Netherlands. He is expected to defend his thesis in the summer of 2026 and is currently working as an internal medicine resident\, after which he will begin an oncology fellowship in 2027. His research focuses on melanoma\, in which he and his colleagues have assembled a multicentre dataset of digitized whole-slide images\, CT scans\, and clinical data\, with the aim of improving prediction of response to first-line immune checkpoint inhibition in patients with advanced melanoma. His work has primarily focused on histopathological analysis\, including tumor-infiltrating lymphocyte (TIL) assessment\, as well as on the organization of the PUMA challenge\, which investigated whether additional immune cell subtypes provide added predictive value.

This has motivated continued interest in engineered image-based biomarkers\, which offer greater interpretability but are often proposed based on anecdotal evidence or fragmented prior literature rather than systematic biological validation. We introduce SAGE (Structured Agentic system for hypothesis Generation and Evaluation)\, an agentic AI system designed to identify interpretable\, engineered pathology biomarkers by grounding them in biological evidence. SAGE integrates literature-anchored reasoning with multi-modal data analysis to correlate image-derived features with molecular biomarkers\, such as gene expression\, and clinically relevant outcomes. By coordinating specialized agents for biological contextualization and empirical hypothesis validation\, SAGE prioritizes transparent\, biologically supported biomarkers and advances the clinical translation of computational pathology. \nRegister NowBio\nDr. Sahar Almahfouz Nasser is a postdoctoral researcher at Emory University’s Madabhushi Lab\, working at the intersection of computational pathology\, agentic AI\, and translational oncology. He also serves as Associate Chief of Radiology\, Associate Chief Medical Information Officer\, and Associate Chief of Staff. Internationally recognized for advancing pediatric radiology\, imaging informatics\, and quality improvement\, he brings particular expertise in abdominal and cancer imaging and in the use of imaging data to improve clinical care and interoperability across the healthcare enterprise. \nA prolific scholar and educator\, Dr. Towbin has delivered invited lectures worldwide and published more than 400 manuscripts and 150 book chapters. He currently serves as Chair-Elect of the Society for Imaging Informatics in Medicine and previously served as co-chair of the HIMSS–SIIM Enterprise Imaging Community. His work bridges clinical care\, research\, and informatics to improve outcomes for children\, families\, and the healthcare workforce. \nKevin Schap\, MScBMI\nDirector of Clinic Informatics\, Digital Pathology\, and Data Science\nCollege of American Pathologists \nKevin Schap\, MScBMI\, is Director of Clinical Informatics\, Digital Pathology\, and Data Science at the College of American Pathologists. Kevin oversees multiple CAP initiatives\, including the AI Committee\, Digital and Computational Pathology Committee\, Informatics Committee\, and the Council on Informatics and Pathology Innovation. SUMMARY:Analysis of computational tumor-infiltrating lymphocytes in breast cancer from the results of the TIGER challenge

The series invites researchers and leaders in computational pathology and related fields to present their work and foster thought-provoking discussions. \nThis seminar features Mart van Rijthoven\, Radboud UMC\, Nijmegen\, The Netherlands. \nAbstract\nTumor-infiltrating lymphocytes (TILs) is a recognized prognostic biomarker in breast cancer. However\, poor interobserver agreement and limited reproducibility highlight the need for computational approaches. Despite advances\, adoption of computational models has been hindered by lack of standardized methods and robust benchmarks. To address this\, we launched TIGER\, an international competition to build open-source computational TILs (cTILs) models. Here\, we present a multi-centric analysis of cTILs methods on resections and biopsies from 3\,708 human epidermal growth factor receptor 2-positive (HER2+) and triple-negative breast cancers (TNBC) from clinical practice and phase 3 trials. We report benchmarks on image analysis performance\, show strong agreement of cTILs with pathologists\, and demonstrate positive association of cTILs with neoadjuvant therapy response in HER2+\, superior to visually scored TILs. We also show that cTILs add independent prognostic information to clinical variables in TNBC resections. He also developed the WholeSlideData package and co-organized the TIGER challenge.
