SUMMARY:TIA Centre Seminar Series – Computational Pathology for Discovering Biomarkers of Immunotherapy Response in Advanced Melanoma
DESCRIPTION:0000Days00Hrs00Min00SecThe Tissue Image Analytics (TIA) Centre hosts a seminar series for those interested in computational pathology. The series invites researchers and leaders in computational pathology and related fields to present their work and foster thought-provoking discussions. \nThis seminar features Dr Mark Schuiveling from UMC Utrecht\, the Netherlands. \nAbstract\nImmune checkpoint inhibition (ICI) has significantly improved outcomes for patients with advanced cutaneous melanoma\, leading to durable responses and potential long-term remission in approximately half of patients. However\, it is also associated with severe and sometimes irreversible toxicity\, underscoring the need for reliable biomarkers. In our group\, we investigate whether computational pathology approaches add value beyond clinical data for identifying biomarkers associated with treatment response in these patients. In this talk\, I will highlight our work on tumor-infiltrating lymphocytes in melanoma histopathology\, as well as our recent work in the PUMA challenge\, where we assessed whether additional immune cell subsets provide further associated predictive value. \nRegister NowBio\nMark Schuiveling is a medical doctor and PhD candidate at the University Medical Centre Utrecht\, the Netherlands. He is expected to defend his thesis in the summer of 2026 and is currently working as an internal medicine resident\, after which he will begin an oncology fellowship in 2027. His research focuses on melanoma\, in which he and his colleagues have assembled a multicentre dataset of digitized whole-slide images\, CT scans\, and clinical data\, with the aim of improving prediction of response to first-line immune checkpoint inhibition in patients with advanced melanoma. His work has primarily focused on histopathological analysis\, including tumor-infiltrating lymphocyte (TIL) assessment\, as well as on the organization of the PUMA challenge\, which investigated whether additional immune cell subtypes provide added predictive value.

DESCRIPTION:0000Days00Hrs00Min00SecOPEN LETTER:\nANIL PARWANI Professor of Pathology and Biomedical Informatics\, Chair of Pathology Department\, The Ohio State University \nDear Colleagues\, \nIt is my great pleasure to offer my full support for the 12th Digital Pathology & AI Congress\, co-located with the 3rd American Spatial Biology Congress\, taking place on May 7–8\, 2026 in Columbus\, Ohio. \nWe are at a defining moment in the evolution of pathology. Digital pathology and artificial intelligence are no longer future concepts—they are actively reshaping how we diagnose disease\, generate knowledge from tissues\, and deliver precision medicine to patients. When combined with advances in spatial biology\, these technologies provide unprecedented insight into tissue architecture\, cellular interactions\, and disease biology across scales. \nAt The Ohio State University\, we have witnessed firsthand how digital pathology and AI can transform clinical workflows\, enable computational discovery\, and bridge research and patient care. From whole-slide imaging and AI-assisted diagnostics to spatial multi-omics and translational research\, these tools are redefining what is possible in modern pathology. Importantly\, their successful adoption depends not only on technological innovation\, but also on collaboration\, shared learning\, and open dialogue across academia\, healthcare\, and industry. \nThis Congress brings together exactly that community. By uniting pathologists\, scientists\, clinicians\, computational experts\, and technology innovators\, the meeting provides a vital forum to explore real-world implementation\, regulatory considerations\, and emerging scientific applications. The co-location of digital pathology and spatial biology is particularly timely\, reflecting how closely these disciplines are converging to drive the next generation of precision diagnostics and therapeutics. \nI am especially pleased that this meeting is being held in Columbus and in collaboration with The Ohio State University. Her work included leading large‑scale implementations such as Radiology PACS systems\, Epic\, and automated chemistry lines—projects that required deep technical expertise\, operational insight\, and cross‑disciplinary coordination. \nOutside of her professional work\, Jennifer is an avid cyclist and runner. \nJonathan Shoemaker \nJonathan Shoemaker is an accomplished healthcare technology leader with nearly 30 years of experience advancing enterprise imaging\, workflow modernization\, and clinical informatics across major academic medical centers. As Executive Director of Imaging Systems and Services at Stanford Health Care\, he oversees a broad imaging portfolio and led the design of a highly resilient enterprise imaging platform that supports AI\, machine learning\, and real‑time research integration. Recently Mr. Shoemaker’s portfolio architected the latest Digital pathology platform that enables a full dicom deployment and informatics normalization model into Stanford digital pathology. \nPreviously\, at Sutter Health\, Jonathan directed one of the nation’s largest deconstructed PACS implementations\, consolidating 54 medical record systems and migrating 2PB of imaging data while building a new enterprise imaging support organization. His earlier consulting and engineering roles—spanning Ascendian Healthcare Consulting and GE Healthcare—shaped his reputation as a national expert in imaging architecture\, VNA strategy\, workflow optimization\, and complex multi‑system integration. \nRebecca Rojansky  \nDr. Rebecca Rojansky\, MD. PhD. is Associate Director for Clinical Pathology Informatics and Assistant Professor of Pathology at Stanford University School of Medicine. She is a practicing molecular and hematopathologist. Her work focuses on digital and computational pathology\, artificial intelligence\, and clinical informatics infrastructure to enable precision diagnostics.

SUMMARY:Analysis of computational tumor-infiltrating lymphocytes in breast cancer from the results of the TIGER challenge

The series invites researchers and leaders in computational pathology and related fields to present their work and foster thought-provoking discussions. \nThis seminar features Mart van Rijthoven\, Radboud UMC\, Nijmegen\, The Netherlands. \nAbstract\nTumor-infiltrating lymphocytes (TILs) is a recognized prognostic biomarker in breast cancer. However\, poor interobserver agreement and limited reproducibility highlight the need for computational approaches. Despite advances\, adoption of computational models has been hindered by lack of standardized methods and robust benchmarks. To address this\, we launched TIGER\, an international competition to build open-source computational TILs (cTILs) models. Here\, we present a multi-centric analysis of cTILs methods on resections and biopsies from 3\,708 human epidermal growth factor receptor 2-positive (HER2+) and triple-negative breast cancers (TNBC) from clinical practice and phase 3 trials. We report benchmarks on image analysis performance\, show strong agreement of cTILs with pathologists\, and demonstrate positive association of cTILs with neoadjuvant therapy response in HER2+\, superior to visually scored TILs. We also show that cTILs add independent prognostic information to clinical variables in TNBC resections. He also developed the WholeSlideData package and co-organized the TIGER challenge.
